The Science
Evidence & Mechanism
MOTS-c is a mitochondria-derived peptide encoded within the mitochondrial 12S rRNA gene. It has been shown in preclinical studies to influence mitochondrial function, activate AMPK signaling, regulate metabolic pathways such as the folate cycle and de novo purine synthesis, and mediate mitochondria-to-nucleus retrograde signaling. These mechanisms make MOTS-c an important subject of ongoing research in metabolic health, cellular function and healthy aging.

MOTS-c is a peptide whose sequence is encoded in mitochondrial DNA. Cell and mouse studies link it to AMPK, a system that helps cells respond to changes in available energy. In mice, researchers reported improved insulin sensitivity and protection against diet-related metabolic changes. These findings explain the interest in glucose regulation, but do not establish a human treatment benefit.
MOTS-c improved exercise performance in young and older mice in a 2021 study. The human part measured naturally occurring MOTS-c around exercise, rather than testing whether administering the peptide improves performance. This is an important distinction: a rise after exercise is a biological observation, while better endurance or recovery requires a controlled human intervention trial.
These compounds address different research questions. MOTS-c is studied in metabolic signalling, SS-31 in interactions with mitochondrial membranes, and 5-Amino-1MQ in inhibition of the NNMT enzyme. Their mechanisms do not show that they are interchangeable or that combining them improves energy, fat loss or longevity. Clinical outcomes must be demonstrated for the exact compound and preparation.
